This is probably one case where that test might not be a bad idea.

Lupron causes hormonal changes, it does not directly attack adrenal
tissue as something like Lysodren would do. But I believe that by
reducing the constant stimulation, hyperplasia may be reduced or
prevented (I hope one the vets out there will bail me out on this
one... I'm definitely in over my head!) though if there is a tumor,
it will continue to grow.

I believe that at least one vet here supports giving Lupron following
adrenal surgery to prevent further stimulation of the remaining
gland. I won't mention a name in case I've misread, misunderstood
something along the way, or if my memory is not that great.

I have not personally used Melatonin, so I can't offer any
information on how useful that may be.
