The earliest I've seen a response to Lupron has been three weeks
after the first shot. With another ferret, it took two or three
weeks after the second shot before I saw a response. This has been
for hair regrowth as they had no other noticeable symptoms. The
length of time the hair follicles have been atrophied may have a
bearing on this too.

I can't help you on your melatonin question.
