Yes, the Lupron shot does hurt. We usually go with surgery because the Lupron shot is not curative. Two of our's have had the shots: one who was elderly and a poor surgical candidate for other reasons, and one who fortunately just needed a hormonal re-set a decent while after surgery (something that was confirmed during much later surgery and by her never needign more Lupron). Surgery cures usually; Luron doesn't.

Two things which have nothing to do with the Lupron shot came to mind from your description.

One is that influenza season has been with us for a few weeks now. I don't know if this is true elsewhere, but it is on the Eastern Seaboard. Ferrets do get influenza. Obviously, influenza is primarily a respiratory disease, but GI upset can happen as well, as we all know.

The second is that it is shedding season and the ingested fur tends to cause throat tickles and vomiting (which reminds me that I haven't combed the ferrets yet today to remove loose fur...).

Of course, it could be a mild reaction to the shot, but keep a placed open for other possibilities.
