There could be a clue here to why the young ferret isn't using the litter
box. For one reason or another, he may not like the tube, especially if it
requires acrobatics for him to get up and down levels. (Imagine trying to
potty train a small human child when the toilet isn't right down the hall,
but instead it's down a few flights of stairs, almost in another
building...) Or he may be fine with the tube when the light is on, but not
like using it in the dark.
My ferrets generally haven't liked those tubes much, especially not when
the tubes require vertical climbing. I took them out and made little
'stairways' out of mesh wire. The tubes are now on the floor, and the
ferrets like to play in them.
And it could be that the ferret isn't interested in using the box. Some
ferrets are a real pain to get litter box trained. I have a 5-year old
female in perfect health who will NOT go down or up to get to a litter
box--so she's in one story cage with no levels. That works pretty well for
both of us.
