I strongly agree with Mike on this.

Remember that infection also causes nodes to enlarge.

Remember, too, that many ferrets put fat around their nodes and at times they do that so well that even vets who treat a lot of ferrets can't tell if they are feeling an enlarged node or fat.

How many nodes are up and by how much? How hard are they?

An aspirate is the easiest way to go, but it does have a higher rate of false positives and false negatives. It's a fast way to find it what is being felt is fat, though. Removal of an affected node is more difficult; it's surgery, and how difficult depends on the node location. It is more accurate.
