The other person that mentioned the 2 mg dose every three to four months may have been talking about the 4-month Lupron Depot, but didn't really say. It seems likely that one wouldn't give 1-month Lupron every three to four months. 2 mg of the 4-month Lupron Depot would equate to 500 mcg per month over a four month period, the same amount that you're currently giving. 500 mcg per month is more than most ferrets need. I don't know if even more than that would help.

Lupron doesn't always work and/or may stop working at some point in time, regardless of the dosage. This could be the case if there was a functional tumor. That is, a tumor that is producing hormones regardless of any outside influence. An insulinoma would be a good example of one of these. It's pumping out insulin which cannot be shut down by the body's normal feedback mechanism. Lupron doesn't act directly on the adrenal glands, but shuts off a couple hormones that the pituitary puts out which in turn stimulate the adrenal glands into producing sex hormones that are the problem. This is the "outside influence" on which Lupron acts. Melatonin performs the same function, but in a slightly different manner.
