If you are talking about the dark stripe I have read suppositions but no one knows.

There are ferrets who get a white stripe on their heads as one of the possible markings of a neural crest genetic variant (often called WS but that is likely the wrong term). Here are some sources of info:

Yes, humans can be allergic to ferrets, though it is not a common allergy. There is even a case of an anaphylactic reaction on record. I think that case may be the one in J Allergy Clin Immunol. 2001 May;107(5):927.

Whole males usually seem to be most commonly mentioned and we personally consider it wise for those who are very allergic to cats to avoid whole males because we have heard of too many with both allergies.
