Yes you can check with a human device but the first time you should
do it at the vets office and have him do the same test so you can
compare your machine with his reading. Human devices read lower than
the actual reading because there is a different red blood cell to
plasma ratio in ferret blood to human blood. When you know how far
out your meter is it is OK do do it at home.
As for the clicker it is normal to do it in a paw pad so the
adjustment will depend on the thickness of the pad, it will be trial
and error but the vet will be there to advise you the first time. If
you massage the ferrets foot first the blood will flow more easily.
