it is my understanding that ferrets spleens get enlarged from time to time is normal ...and it is ok per a vet i saw.
Of course he should be checked out to see if there it is health related

[Older ferrets often have enlarged spleens, but there are
limits to what is safe and acceptable. That may be in
response to long term suppressed infection, for example,
of the species of Helicobacter that ferrets get (not the
same species as humans get).

Spleens will enlarge with infections and will enlarge with
lymphoma. Those are the two most common causes
though not the only causes, for example, pancreatic
carcinoma can blow up splenic size and so can other things.
