> In epidemiologic studies, high intake of =CE=B2-cryptoxanthin has been as=
sociated with a decreased risk of lung cancer, particularly among current s=
mokers. However, data are not available from well-controlled animal studies=
to examine the effects of =CE=B2-cryptoxanthin on cigarette smoke-induced =
lung lesions, and the biological mechanisms by which =CE=B2-cryptoxanthin m=
ight affect lung carcinogenesis. We evaluated the effects of =CE=B2-cryptox=
anthin supplementation on cigarette smoke-induced squamous metaplasia, infl=
ammation, and changes in protein levels of pro-inflammatory cytokine [tumor=
necrosis factor alpha (TNF=CE=B1)] and transcription factors [nuclear fact=
or kappa B (NF-=CE=BAB) and activator protein-1 (AP-1)], as well as on smok=
e-induced oxidative DNA damage [8-hydroxy-2'-deoxyguanosine (8-OHdG)] in th=
e lung tissue of ferrets. Thirty six male ferrets were assigned to cigarett=
e smoke exposure or no exposure and to low-dose, or high-dose =CE=B2-crypto=
xanthin, or no dose (2 x 3 factorial design) for 3 months. =CE=B2-Cryptoxan=
thin supplementation dose-dependently increased plasma and lung =CE=B2-cryp=
toxanthin levels in ferrets, whereas cigarette smoke exposure lowered plasm=
a and lung =CE=B2-cryptoxanthin levels. =CE=B2-Cryptoxanthin at both doses =
significantly decreased smoke-induced lung squamous metaplasia and inflamma=
tion. =CE=B2-Cryptoxanthin also substantially reduced smoke-elevated TNF=CE=
=B1 levels in alveolar, bronchial, bronchiolar and bronchial serous/mucous =
gland epithelial cells and in lung macrophages. Moreover, =CE=B2-cryptoxant=
hin decreased smoke-induced activation of NF-=CE=BAB, expression of AP-1 an=
d levels of 8-OHdG. The beneficial effects of =CE=B2-cryptoxanthin were str=
onger for high-dose =CE=B2-cryptoxanthin than for low-dose =CE=B2-cryptoxan=
thin. Data from this study indicate that =CE=B2-cryptoxanthin provides a be=
neficial effect against cigarette smoke-induced inflammation, oxidative DNA=
damage and squamous metaplasia in the lungs.
